Hey Guys beyond the coronavirus cases, I have been watching the new bills coming out.

Of particular interest is the new bill, coronavirus stimulus package (link below)

What interests me is the below

PAYCHECK PROTECTION PROGRAM: Provides 8 weeks of cash flow assistance up to $10 million to small businesses through 100% federally guaranteed no and low-interest loans that are eligible for forgiveness, under circumstances described below under “loan forgiveness.” Funds may be used for payroll wages and benefits, rent, mortgage payments and utilities, all of which is eligible for loan forgiveness.

LOAN FORGIVENESS: Distributions of federally-guaranteed small business loans that are used for payrolls that include employee salaries, cash tips, paid sick leave, insurance premiums, mortgage payments, utilities, and other debt obligations will be forgiven.

https://finance.yahoo.com/news/coronavirus-stimulus-package-aid-and-stimulus-guide-for-us-individuals-and-businesses-223030400.html

So I am trying to figure this out, but are they saying if say a small business has a payroll of 20K in the next 8 weeks..that business is allowed to borrow a loan for 20K..but that loan is forgiven! As in the government is giving small businesses money to meet needs like payroll and the like?

And if that is the case, would this mean a lot of businesses wouldn't lay off their workers..and I also read if you rehired workers you layed off, you can still the money for this purpose.

finally had a chance to listen to white house press conference today.
for all the people who still deny the threat of covid19, it was pretty grim today.

fauci and birx just predicted 80,000 to 200,000 dead even with all the social distancing.
without the social distancing, they said 1-2 million would have died.
so the guidelines will be extended until end of april.

for those who insist covid19 is just a flu, perhaps they could explain what is the mysterious bug that would cause 1-2 million fatal cases of pneumonia...

I think those numbers were put out there to scare people, but anyone who thinks this is just a flu needs to get their brain checked. Even trump changed his tone

I wouldn't know why anyone wouldn't deny the threat of covid 19...but I guess they exists out there.

The flu is different..perhaps a long time ago, the flu had taken more people..but with the flu vaccine, the flu can't spread as easily because there are too many people immune. It would have to catch onto someone that isn't immune than that person have to transmit it to someone else that isn't immune...not likely..making it harder to spread

There have been years where the flu vaccine has gotten the wrong type of flu virus and thus had smaller types of epidemics..but even thing, people still retain some type of immunity.

Someday the same thing maybe with Covid-19 once there is vaccine..but for now, that isn't the case

yes, to my surprise, trump tried to show his 'sensitive' side today.

Very grim today. I am still shocked that these updates are not nationally televised by everyone. I think the DR's stopped short of saying people are idiots for not being socially responsible (St. Pat's, Spring Break, Mardi Gras, failure to self quarantine, etc.). Some people are self absorbed douche bags and are clearly risking lives by their ignorance.

thank you for pointing out the 'some type of immunity' even when the vaccine makers guess incorrectly re the type of virus for the year. some do not get that.

actually, if actemra or other il6 blocker works on covid19, there is a reasonable chance that it would be useful for other pneumonia cases. since 'normal' pneumonia is a top-10 killer, perhaps a lot of those lives could be saved given the huge efforts put towards covid19.

on my daily walk today, i learned from some neighbors that a decently large group of cars were parked on a nearby street for a party yesterday. then they told me that someone called the police who gave all the cars tickets.

all 3 of us hope that the tickets were at least $1k.

This whole process has just made me value truthfulness. This pandemic is unprecedented. And that uncertainty by itself is enough worry. Don't need to be second guessing if what we're hearing is an overestimate, underestimate, fact, or fiction on top of that.

Others have already commented but this is badly misleading. I'm directly in touch with a hospital in Brooklyn NY that's squeezed in nearly thrice (yes thrice) it's capacity of patients as we speak and is desperate for protective gear, supplies and equipment. Granted it's one hospital in a hot-spot, but still.

kevzara trials have actually just started outside usa. kevzara is similar to actemra in the sense that they block il6 pathway to reduce inflammation. but one goes after the il6 receptor while the other goes after il6 itself.

https://www.pmlive.com/pharma_news/sanofi_doses_first_patient_in_global_kevzara_covid-19_trial_1331903

"The first patient outside the US has been treated with Kevzara in Sanofi and Regeneron’s global clinical trial programme, which is evaluating the drug in severely ill COVID-19 patients.

Sanofi, which is leading the Kevzara (sarilumab) trials outside the US, announced that the first patient had been dosed today, as the phase 2/3 trial is initiated in Italy, Spain, Germany, France, Canada and Russia."

"Kevzara – an interleukin-6 inhibitor – is being evaluated on the basis that IL-6 may play a role in driving the overactive inflammatory response in the lungs of critically and severely ill COVID-19 patients.

This is supported by initial results from a study in China that evaluated another IL-6 inhibitor – Roche’s Actemra (tocilizumab) in severely ill COVID-19 patients.

This small-scale study demonstrated that treatment with the IL-6 inhibitor caused COVID-19 patients to experience rapidly reduced fevers, and 75% of patients reduced their need for supplemental oxygen within days of receiving the drug.

As a result of this preliminary data, Roche won an approval in China for Actemra as a treatment for patients with severe or critical COVID-19."

NBA Program Asking Players Who Recovered From COVID-19 To Donate Plasma For Research

Quote:
The NBA, through its "NBA Together" program, is asking players and team individuals who have recovered from COVID-19 to donate their plasma as research.

The donation would help support the National COVID-19 Convalescent Plasma Project, which features top specialists exploring plasma as a potential treatment for the coronavirus.

The NBA recently launched the "NBA Together" program to help educate and engage audiences during the COVID-19 pandemic.

The campaign hopes to amplify information and resources to keep communities connected throughout the pandemic.

i was working on a biomed collaboration project in china for 6 months and of course this puts a stop to everything for a long time. same for another sensor project which now cannot raise funding with an ai partner in china.

commerce outside a few things has stopped for everyone.

johnson & johnson announces several vaccine candidates. their vaccine is probably a more traditional type unlike the mRNA vaccine that went into safety trials a few days ago. their timeline is phase 1 trials latest by september. they are accelerating production so vaccine available for emergency use by early 2021. in comparison, typical vaccine development is 5-7 years.

https://www.jnj.com/johnson-johnson-announces-a-lead-vaccine-candidate-for-covid-19-landmark-new-partnership-with-u-s-department-of-health-human-services-and-commitment-to-supply-one-billion-vaccines-worldwide-for-emergency-pandemic-use

"Johnson & Johnson Announces a Lead Vaccine Candidate for COVID-19; Landmark New Partnership with U.S. Department of Health & Human Services; and Commitment to Supply One Billion Vaccines Worldwide for Emergency Pandemic Use
Johnson & Johnson and BARDA Together Commit More than $1 Billion to Novel Coronavirus Vaccine Research and Development; Company Expects to Initiate Phase 1 Human Clinical Studies of Vaccine Candidate at Latest by September 2020
Johnson & Johnson Will Establish New U.S. Vaccine Manufacturing Capabilities and Additional Production Capacity Outside the U.S. to Begin Production at Risk to Help Ensure Global Vaccine Supply
NEW BRUNSWICK, N.J., March 30, 2020 – Johnson & Johnson (NYSE: JNJ) (the Company) today announced the selection of a lead COVID-19 vaccine candidate from constructs it has been working on since January 2020; the significant expansion of the existing partnership between the Janssen Pharmaceutical Companies of Johnson & Johnson and the Biomedical Advanced Research and Development Authority (BARDA); and the rapid scaling of the Company’s manufacturing capacity with the goal of providing global supply of more than one billion doses of a vaccine. The Company expects to initiate human clinical studies of its lead vaccine candidate at the latest by September 2020 and anticipates the first batches of a COVID-19 vaccine could be available for emergency use authorization in early 2021, a substantially accelerated timeframe in comparison to the typical vaccine development process.

Through a landmark new partnership, BARDA, which is part of the Office of the Assistant Secretary for Preparedness and Response (ASPR) at the U.S. Department of Health and Human Services, and Johnson & Johnson together have committed more than $1 billion of investment to co-fund vaccine research, development, and clinical testing. Johnson & Johnson will use its validated vaccine platform and is allocating resources, including personnel and infrastructure globally, as needed, to focus on these efforts. Separately, BARDA and the Company have provided additional funding that will enable expansion of their ongoing work to identify potential antiviral treatments against the novel coronavirus.

As part of its commitment, Johnson & Johnson is also expanding the Company’s global manufacturing capacity, including through the establishment of new U.S. vaccine manufacturing capabilities and scaling up capacity in other countries. The additional capacity will assist in the rapid production of a vaccine and will enable the supply of more than one billion doses of a safe and effective vaccine globally. The Company plans to begin production at risk imminently and is committed to bringing an affordable vaccine to the public on a not-for-profit basis for emergency pandemic use.

Alex Gorsky, Chairman and Chief Executive Officer, Johnson & Johnson, said, “The world is facing an urgent public health crisis and we are committed to doing our part to make a COVID-19 vaccine available and affordable globally as quickly as possible. As the world’s largest healthcare company, we feel a deep responsibility to improve the health of people around the world every day. Johnson & Johnson is well positioned through our combination of scientific expertise, operational scale and financial strength to bring our resources in collaboration with others to accelerate the fight against this pandemic.”

Paul Stoffels, M.D., Vice Chairman of the Executive Committee and Chief Scientific Officer, Johnson & Johnson, said, “We greatly value the U.S. government’s confidence and support for our R&D efforts. Johnson & Johnson’s global team of experts has ramped up our research and development processes to unprecedented levels, and our teams are working tirelessly alongside BARDA, scientific partners, and global health authorities. We are very pleased to have identified a lead vaccine candidate from the constructs we have been working on since January. We are moving on an accelerated timeline toward Phase 1 human clinical trials at the latest by September 2020 and, supported by the global production capability that we are scaling up in parallel to this testing, we expect a vaccine could be ready for emergency use in early 2021.”

Johnson & Johnson’s Lead COVID-19 Vaccine Candidate
Johnson & Johnson began efforts in January 2020, as soon as the novel coronavirus (COVID-19) sequence became available, to research potential vaccine candidates. Research teams at Janssen, in collaboration with Beth Israel Deaconess Medical Center, part of Harvard Medical School, constructed and tested multiple vaccine candidates using the Janssen AdVac® technology.

Through collaborations with scientists at multiple academic institutions, the vaccine constructs were then tested to identify those with the most promise in producing an immune response in preclinical testing.

Based on this work, Johnson & Johnson has identified a lead COVID-19 vaccine candidate (with two back-ups), which will progress into the first manufacturing steps. Under an accelerated timeline, the Company is aiming to initiate a Phase 1 clinical study in September 2020, with clinical data on safety and efficacy expected to be available by the end of the year. This could allow vaccine availability for emergency use in early 2021. For comparison, the typical vaccine development process involves a number of different research stages, spanning 5 to 7 years, before a candidate is even considered for approval.

For more than 20 years, Johnson & Johnson has invested billions of dollars in antivirals and vaccine capabilities. The COVID-19 vaccine program is leveraging Janssen’s proven AdVac® and PER.C6® technologies that provide the ability to rapidly develop new vaccine candidates and upscale production of the optimal vaccine candidate. The same technology was used to develop and manufacture the Company’s Ebola vaccine and construct our Zika, RSV, and HIV vaccine candidates which are in Phase 2 or Phase 3 clinical development stages."

One of my sons is in a terrible state of depression because of how this crisis came along at the worst possible moment for him, with the worst possible consequences including sheltering in place, social isolation and closing of businesses and events. Terrible timing for him, to an uncanny degree. "Hang in there" doesn't cut if for him. The best I can do for my son is be there for him. The rays of hope I see are the breadth of trials getting started on treatments and hopefully a bit of bridging of the financial gaps from the stimulus package. If anything is going to bring this to a close more quickly than letting it burn through the population unchecked killing millions, it's these clinical trials getting started all over the world. I know a chemical physicist involved in this effort, there are some brilliant people involved, very dedicated to the science and the urgency. There will come a time when we can pick up the pieces and move again. Not sure if this helps at all, but you are not alone in this.

if one of the il6 drugs work, then they just need more ventillators.

of course, given the choice, i would take an antiviral like chloroquine/remdesivir working as that could stop the progression from moderate/severe cases to critical cases. chloroquine would be the best as there is the most of it.

Getting back to basketball...I'm warming to the idea of having all play-off teams quarantined in Las Vegas and having the play-offs there. It will definitely be awkward with no fans...It'll be like a pick up game with a lot more at stake. Nonetheless, it beats the alternative of no basketball IMO.

that could work, though i am not sure the player would be willing to be jailed for 2 weeks.

hope this works because the whole world could use a distraction.

yet more on IL6 inhibitors with a little info on trials. in particular, actemra/tocilizumab is actually fda approved to manage cytokline storms for a different condition.

"In an editorial, researchers from the Society for Immunotherapy of Cancer (SITC) suggested that though definitive data showing that IL-6 receptor blockade benefits patients with COVID-19 induced pneumonitis are currently lacking, an effort should be made to maximize the availability of anti-IL-6 agents on a compassionate basis to treat hospitalized critically ill patients infected with COVID-19."

"The use of IL-6 or IL-6 receptor blocking antibodies like tocilizumab (Actemra), sarilumab (Kevzara), and siltuximab (Sylvant) that are FDA approved for various conditions including rheumatologic disease and the lymphoproliferative disorder Castleman’s syndrome could potentially be used to treated hospitalized critically ill patients with COVID-19-induced hypoxia, according to the researchers."

"Tocilizumab specifically has data from the frontlines of the pandemic that indicate that the agent might offer lifesaving benefit for patients with COVID-19 in respiratory distress. Currently, tocilizumab is FDA approved to manage cytokine release syndrome (CRS) in patients receiving CAR T-cell therapy."

"However, tocilizumab has been shown to reduce toxicity in patients treated with immune checkpoint inhibitors who were steroid refractory and has been added to ipilimumab (Yervoy) and nivolumab (Opdivo) in an ongoing phase II study. In Castleman’s disease, the agent has been shown to reduce viral loads. Moreover, tocilizumab is being explored as a possible supportive care measure for the management of CRS in patients with cancer treated with several CD3-based bispecific molecules."

"The researchers suggested straightforward parameters for the implementation of anti-IL-6 agents, including that complete blood counts and differentials, serum LDH, ferritin, CRP, and IL-6 be recorded in treated patients, that serum be retained for future evaluation, and simple clinical parameters be assessed including time in ICU, days of hospitalization, and pulmonary parameters including FEV1, Fi02, PaO2/FiO2 ratio and oxygen need be recorded.

Investigators in Italy and China who have used tocilizumab to treat patients infected with COVID-19 at doses comparable to those used for the management of CRS have reported rapid improvement in both intubated and non-intubated patients. In these reports, the expedited administration of anti-IL-6R therapy for patients in acute respiratory distress has been essential.

A recent study protocol to evaluate the efficacy of tocilizumab in COVID-19 induced pneumonitis was able to accrue over 300 patients worldwide in less than 24 hours. Genentech also indicated that they will provide 10,000 vials of the agent to the US Strategic National Stockpile. In China, tocilizumab was approved in March 2020 for the treatment of patients infected with COVID-19 who experienced serious lung damage and elevated IL-6.

In the US, a trial of sarilumab in the COVID-19 setting remains ongoing."
